ABSTRACT

C18 ceramide plays an important role in the occurrence and development of oral squamous cell carcinoma. However, the function of ceramide synthase 1, a key enzyme in C18 ceramide synthesis, in oral squamous cell carcinoma is still unclear. The aim of our study was to investigate the relationship between ceramide synthase 1 and oral cancer. In this study, we found that the expression of ceramide synthase 1 was downregulated in oral cancer tissues and cell lines. In a mouse oral squamous cell carcinoma model induced by 4-nitroquinolin-1-oxide, ceramide synthase 1 knockout was associated with the severity of oral malignant transformation. Immunohistochemical studies showed significant upregulation of PCNA, MMP2, MMP9, and BCL2 expression and downregulation of BAX expression in the pathological hyperplastic area. In addition, ceramide synthase 1 knockdown promoted cell proliferation, migration, and invasion in vitro. Overexpression of CERS1 obtained the opposite effect. Ceramide synthase 1 knockdown caused endoplasmic reticulum stress and induced the VEGFA upregulation. Activating transcription factor 4 is responsible for ceramide synthase 1 knockdown caused VEGFA transcriptional upregulation. In addition, mild endoplasmic reticulum stress caused by ceramide synthase 1 knockdown could induce cisplatin resistance. Taken together, our study suggests that ceramide synthase 1 is downregulated in oral cancer and promotes the aggressiveness of oral squamous cell carcinoma and chemotherapeutic drug resistance.

ABSTRACT

Objective:To explore the therapeutic effect of cistanche deserticola ethanol extraction (CDE) in the rats with osteoporosis induced by ovariectomy,and to clarify its mechanism.Methods:A total of 72 female SD rats were randomly divided into control,model,estradiol (0.3 g · kg-1),low,middle and high doses (0.5,1.0 and 2.0 mg · kg-1) of CDE groups,and 12 rats in each group.The rat ovaries were removed by operation in the other groups to establish the osteoporosis models,while the same size of fat of the rats in control group was excised.5 d later,the rats in estradiol group were hypodermicly injected with estradiol,and the rats in CDE groups were orally administrated with corresponding doses of CDE.The same volume of diluted water was given to the rats in control and model groups.After 20 weeks,the density of right femur of the rat was detected with the method of ratio of weight and size;the mechanics index of left femur of the rat was analyzed by bone granulometer;the levels of serum alkaline phosphatase (ALP) and osteocalcin were measured by ELISA;the levels of serum Ca2+ and P were detected by automatic biochemical analyzer;the morphology of uterus tissue was observed by HE staining.Results:Compared with model group,the densities of right thighbone of the rats in estradiol group and low,middle and high doses of CDE groups were significantly increased (P<0.05 or P<0.01);there were no differences between low,middle and high doses of CDE groups (P>0.05).Compared with model group and low dose of CDE group,the maximum bending forces,the maximun strains,and the bone rigid coefficients in middle and high doses of CDE groups were remarkably increased (P< 0.05),while the levels of ALP were decreased (P< 0.05).Compared with model group,and low and middle doses of CDE groups,the level of osteocalcin in high dose of CDE group was significantly decreased (P<0.05).Compared with model group,the levels of Ca2+ in serum of the rats in low,middle and high doses of CDE groups were significantly increased (P<0.05);there were no significant differences between low,middle and high doses of CDE groups (P>0.05).Compared with model group,the morphology of uterus tissue of the rats in low,middle and high doses of CDE groups was improved at different degrees.Conclusion:CDE has therapeutic effect on osteoporosis through increasing the levels of serum ALP,osteocalcin and Ca2+ of the rats.

ABSTRACT

Objective:To compare the detection results of histopathology and MRI, CT of liposarcoma of extremities,and to provide guide for increasing the accuracy rate of clinical diagnosis.Methods:The data of 43 cases of liposarcoma of the extremities were collected.MRI plain scan and enhanced CT were used in 18 patients before operation.Sixteen cases were diagnosed by CT plain scan and enhanced scan before operation.Nine cases were diagnosed by MRI and CT plain scan and enhanced scan before operation.All the patients underwent the operation. The intraoperative pathological examination was performed in the lesion tissue.The diagnosed results were assessed according to the classification (high differentiated liposarcoma, mucoid liposarcoma, toliposarcoma and mixed liposarcoma).Results:In all the 43 cases,the upper limb lesion was found in 13 patients and the lower limb lesion was found in 30 patients.The average maximal diameter of the lesions was (14.6±6.1)cm.The fat cells were found in all the patients,with the pathological characteristics of adipocytes and lipopolysaccharide mixture exists, and accompanied by deep-stained pleomorphic cells;the CT examination results showed that the low density shadow with clear boundary and linear separated shadow after CT enhanced;the MRI image showed short signal intensity on T1 weighted imaging and long signal intensity on T2 long signal and short time reversal recovery sequence imaging and other signals;the mass existed the cord-like,linear shadow;the lesion surrounding nerve and blood vessels were squeezed,but the bone structure was relatively complete.The CT and MRI results of well-differentiated lipsarcome showed the fat signal,and the enhanced results showed the mild strengthening.The CT and MRI results of mucinous liposarcoma showed the cystic density and signal, and the enhanced results showed the clound-like orlatticed mild and moderate strengthening;the CT and MRI results of mixed liposarcoma showed the mixed imaging findings of above two types.Conclusion:The pathological examination can provide the exact basis for the diagnosis of type of liposarcoma;CT and MRI imagings can provide basis for j udging the size of the tumor;MRI enhanced scan has an important role in judging tumor border.Combination of three kinds of detection methods can provide the basis for the diagnosis of the disease.

ABSTRACT

Objective:To explore the effect of laparoscopic minimally invasive on anorectal dynamics and serum carcino-embryonic antigen (CEA) and carbohydrate antigen 724 (CA724) in patients with rectal cancer.Methods:38 cases of rectal cancer patients were selected as the research group from March 2014 to March 2016 and 40 cases of non neoplastic intestinal polyps as control group,Preoperative anorectal dynamics index (anal resting pressure (ARP),rectal resting pressure (RRP)and anal maximal contraction pressure (MSP),rectal maximum tolerated volume (MTV) of two groups were compared,Preoperative and 3 d,1,2 weeks after operation of serum CEA and CA724 levels were detected in the research group and the clinical curative effect was observed.Results:There was no significant difference of ARP,RRP,MSP,MTV before operation in research group compared with the control group(P>0.05),and was significantly reduced 2,4 weeks after operation(P<0.05),then gradually restored,and basically recovered to the preoperative level 12 weeks after operation.There was no significant difference of serum CEA and CA724 levels 3 d after operation compared with preoperative in research group (P>0.05),and were significantly lower 1,2 weeks after operation compared with preoperative (P<0.05).The clinical effective rate was 65.8%.Conclusion:Efficacy of laparoscopic minimally invasive in treatment of rectal cancer is effective,can effectively reduce the levels of tumor markers,although there are some influence on anorectal dynamics,but it can be returned to normal in the short term.

ABSTRACT

ABSTRACT:Objective To evaluate the expressions of cell adhesion molecules CD146 and transferrin receptor 1 (TfR1)in ovarian epithelial cancer and investigate their relationship with clinical pathological features of patients with ovarian epithelial cancer (OEC ) so as to further explore the pathogenesis of OEC. Methods Immunohistochemistry was employed to determine the expressions of CD146 and TfR1 in normal,benign and ovarian cancer tissues.Results The immunohistochemical results showed that the positive expressions of CD146 and TfR1 gradually increased with the changes of normal,benign and malignant tissues in OEC.There were significant differences between all the groups (P0.05).There was a significant correlation between expressions of CD146 and TfR1 in OEC (P<0.05,rs=0.532).Combined detection sensitivity and specificity were 82.4% and 75.0%.Conclusion The high expressions of CD146 and TfR1 may play a key role in the occurrance,progression and metastasis of OEC.They may be used as potential markers for diagnosis,postoperative follow-up and targeted therapy.

ABSTRACT

Objective To study the expression of insulin like growth factor binding proteins 3 (IGFBP-3) during inhi-bition of resveratrol (Res) on cell proliferation. Methods The inhibitory effect of Res on BGC-823 cells was determined by MTT method; Real-time qRT-PCR and western blot were applied to detect the expression of IGFBP-3 in Res-treated BGC-823 cells. In addition, cytometry was used to determine the proliferation and apoptosis of Res-treated BGC-823 after knockdown of IG-FBP-3 by siRNA. Results Upon Res (20,40, 80 and 160 μmol · L-1 ) treatment,the viability of BGC-823 cells was (82. 35±10. 65)% ,(74. 30±12. 36)% ,(62. 80±14. 66)% and (50. 75±11. 14)% , respectively. The mRNA and protein ex-pression of IGFBP-3 elevated as high as 2. 96-fold compared to the control group (P<0. 05). The cell viability of BGC-823 cells with IGFBP-3 knockdown was significantly higher than that of the wild type ( P < 0. 05 ) only at high Res concentration (160 μmol·L-1 ). Meanwhile,IGFBP-3 knockdown led to a significant decrease on cell apoptotic rate by Res (160 μmol·L-1 ) [(20. 13±9. 12)% vs (35. 48±11. 12)% ,P<0. 05)]. Conclusion Res can inhibit BGC-823 cell proliferation and promote cell apoptosis, the underlying mechanism of which may be related to the overexpression of IGFBP-3 in BGC-823 cells.

ABSTRACT

<p><b>OBJECTIVE</b>To assess the diagnostic efficacy of contrast-enhanced CT in diagnosing mandibular invasion of oral cancers.</p><p><b>METHODS</b>PubMed, Excerpta Medica Database (EMBASE), the European Grey Literature Database, Chinese Biomedical Literature Database (CBM), and CQVIP were searched electronically on January 03, 2014. Hand-searching was also performed on 19 relevant Chinese journals. The methodological quality assessment, as suggested in Cochrane Handbook, and the data extraction of the studies included were performed by two reviewers in duplicate. Meta-analysis was conducted using the Meta Disc 1.4 software.</p><p><b>RESULTS</b>Fourteen studies that involved 642 participants were included. Among the seven prospective studies, one had a low bias risk and the rest had unclear bias risks. When mandibular invasion was diagnosed the Meta-analysis results showed that the pooled sensitivity (SEN) was 0.718 [95% confidence interval (CI), 0.665-0.767], the pooled specificity (SPE) was 0.909 (95% CI, 0.872-0.938), the area under curve (AUC) was 0.906 1, and the Q* was 0.837 8. When mandibular medullary invasion was diagnosed, the SEN was 0.787 (0.643-0.893), the SPE was 0.904 (0.790-0.968), the AUC was 0.949 6, and the Q* was 0.890 0. The Meta-regression results showed that the smaller slide thickness had a higher diagnosis efficacy.</p><p><b>CONCLUSION</b>CT showed a high efficacy in diagnosing mandibular or medullary invasion caused by oral cancers. With a high SPE, contrast-enhanced CT can confirm the clinical diagnosis of bone invasion effectively. In addition, selecting a smaller slide thickness CT can increase diagnosis efficacy.</p>

ABSTRACT

Lateral thoracic flap is a free flap for the reconstruction of soft tissue defects and has been previously used in oral and maxillofacial surgeries. We reported a case involving a huge soft tissue defect on the parotid region caused by the resection of parotid epithelial myoepithelial carcinoma. We discussed the anatomy and application of the lateral thoracic flap.

OBJECTIVE@#To investigate the relationship between tumor-associated carbohydrate antigen sTn and endometrial carcinoma, and to evaluate the diagnostic value of 2 test methods.@*METHODS@#A total of 200 patients were enrolled, including 100 subjects with endometrial carcinoma, 42 healthy nonpregnant women, 15 pregnant women without complications, and 43 patients with benign gynecologic diseases. The serum sTn-antigen concentrations were determined by 2 test methods (3P9 combined with 4A6, and B72.3 combined with CC49).@*RESULTS@#There was a significant difference in the value and the positive rate of sTn in the serum between the subjects and the contrasts (P<0.05). The sTn level in the pregnant women was high. The sTn level in the serum and its positive rate in endometrial carcinoma became higher with the clinical stage. 3P9 combined with 4A6 was better than B72.3 combined with CC49 in the detection of sTn in the serum as to sensitivity, specificity, positive-prediction, negative-prediction, and accuracy.@*CONCLUSION@#The sTn antigen may become a new serological marker for the diagnosis of endometrial carcinoma, but pregnant women should be excluded. 3P9 combined with 4A6 is better than B72.3 combined with CC49 in the detection of sTn in the serum.
